Stunning new visuals of the brain reveal a deceptively simple pattern of organization in the wiring of this complex organ.

Instead of nerve fibers travelling willy-nilly through the brain like spaghetti, as some imaging has suggested, the new portraits reveal two-dimensional sheets of parallel fibers crisscrossing other sheets at right angles in a gridlike structure that folds and contorts with the convolutions of the brain.

This same pattern appeared in the brains of humans, rhesus monkeys, owl monkeys, marmosets and galagos, researchers report Thursday in the journal Science.

"The upshot is the fibers of the brain form a 3-D grid and are organized in this exceptionally simple way," study leader Van Wedeen, a neuroscientist at Harvard Medical School and Massachusetts General Hospital, told LiveScience. "This motif of crossing in three axes is the basic motif of brain tissue." [Inside the Brain: A Journey Through Time]

The organized brain
The surface of the brain contains about 40 billion nerve cells, each making about 1,000 connections in a pattern that brain researchers have yet to decipher, said Marsel Mesulam, the director of the Cognitive Neurology and Alzheimer's Disease Center at Northwestern University. Mesulam, who was not involved in the study, called Wedeen's work "very exciting."

"There can be no more fundamental question in philosophy, in psychology," Mesulam told LiveScience. "The human brain is the single most complex device in the known universe, and it works by nerve cells talking to each other. If we can't figure out how they decide who to talk to and what they tell each other, we just don't understand how the brain functions."

Using a technique he developed called diffusion spectrum magnetic resonance imaging (MRI), Wedeen traced the movement of water molecules along the intersections of brain fibers (the cellular projections that form the brain's communication network), tracking the orientation of each fiber at each crossing.

"What emerged was astonishing," Wedeen said. "What emerged was that the set of fibers that crossed a given fiber, invariably ? and that's a really strong invariably ? look like mutually parallel fibers all coming in like the teeth of a comb and crossing it in one direction." Animal studies had suggested this pattern might exist, and researchers already knew that the nerve cells in the spinal cord and brain stem were organized in very structured parallels and perpendiculars even in humans (consider the long nerve fibers that run down the backbone and then branch out perpendicularly from the vertebrae). But it's difficult to get high-resolution scans of fiber connectivity in the human cortex, given that humans tend to become uncomfortable if left in an MRI scanner for more than 45 minutes or so, Wedeen said. For that reason, images of human brain connections have tended to look like tangled spaghetti, he said.

Wedeen and his colleagues scanned four types of primate brains from deceased animals, enabling them to image the brains for up to 48 hours, as well as brains from living human subjects using a new scanner that can achieve 10 times the resolution of conventional MRI machines. Using special software, the researchers then reconstructed three-dimensional images of the brain-fiber pathways.

"Looking across multiple species, it emerged that the pattern was substantially similar," Wedeen said. "When you went from primates with small brains to primates with big brains ? the rules were the same, but they were being applied more diversely and with more layers in the larger, more complex brains."

Adaptable brain
The finding of clear up-down, front-back and side-to-side organization in the brain makes sense, Wedeen said, given that the brain has had to rewire both evolutionarily (to form the specialized brains humans boast today) and during its lifetime (as it grows and learns, for example). If the organization of communication were chaotic, that wouldn't work.

"It's like rewiring your basement at random," Wedeen said. "First thing that happens, house burns down, you die."

In other words, adapting a complexly wired brain that will still allow the next generation to survive would be next to impossible.

"If you try to picture what would happen if you tried to turn one spaghetti brain into a different spaghetti brain, you realize you would need an impossibly knowledgably intelligent designer standing above the brain and rewiring it," Wedeen said.

With an organized grid structure, however, evolution can easily build on what came before ? adding in a more complex forebrain in humans versus our monkey relatives, for example.

More work should be done to link the imaging methods of Wedeen with traditional neuroanatomy methods to confirm the findings, Mesulam said. Wedeen plans to expand the map of the human brain into more detail. It's also important to understand the relationship between a brain's structure and its function, he said. Understanding the structure of a typical brain would ultimately help scientists comprehend what happens when brain development goes wrong, as in Alzheimer's or mental illness.

"Say somebody comes to you with their 2-year-old and they say, 'My 2-year-old is just not looking me in the eyes.' Is this the first sign of Asperger's or just an individual difference?" Wedeen said. "You'd know how to begin. You'd know what you were doing."

Motorola announces the MOTOACTV in Chile

MotoACTV

While the MotoACTV may have been released in some parts of the world a few months ago, Motorola is continuing their rollout of the device to various parts of the world. Today Motorola has announced they will be bringing their fitness band to Chile next. The MotoACTV has recently been updated to bring some additional features to the unit, all of which will now be able to be enjoyed in even more parts of the world. Full press release after the break.

For most of American history, early spring meant a feast of shad. That tradition has faded, but young chefs are trying to slip the ritual back onto plates.

The earliest Americans from from Florida to Nova Scotia caught shad by the basketful as they swam back from the sea to spawn in their home rivers. The fresh, silvery fish was most certainly a delight after winter's dreary fare. The American shad's Latin name is clue to its allure: Alosa sapadissima, or most delicious herring.

American shad were a key food source for American Indians and later, colonists. New Yorker writer John McPhee called shad the "founding fish" because it fed so many key figures in American history, from William Penn to Abraham Lincoln.

After his presidency, George Washington made a good income from selling shad netted from the Potomac River at Mount Vernon. In 1772, he caught upwards of 1 million shad and herring, according to a charming account of shad's American history by CleanFish founder Dale Sims. Civil War soldiers had rations of shad. And even now, a shad planking is a ritual meal for Virginia politicians.

There are shad on the West Coast, too, most notably in the Columbia River. But they're mere interlopers from the East Coast, having been transplanted there in the 1880s.

In its native rivers, the noble American shad fell on hard times starting in the late 1800s, with overfishing, pollution and dams reducing its numbers to the point where the tradition of shad as the must-have dish for spring had largely faded from memory.

But young chefs are now angling to restore the shad to its former glory. "Since the beginning of people inhabiting this area, there was always shad the first thing in spring, the first guarantee of fresh fish," says Jed Fox, a chef at Ris restaurant in Washington, D.C. Some of those fish would have spawned in Rock Creek, just a few blocks from the restaurant.

It's yummy, and it's local, but is it sustainable? That's a good question.

Many shad fisheries are closed to give the population a chance to rebuild. The Monterey Bay Aquarium Seafood Watch recommends that people avoid eating shad because of that.

But Georgia and South Carolina already have approved plans in place for sustainable commercial shad fishing, and North Carolina has applied for approval.

East Coast shad are managed by the Atlantic States Marine Fisheries Commission, which requires states to have a sustainable fishery plan for American shad in place by Jan. 1, 2013, if they want to continue recreational or commercial fishing. "We recognize the importance of continuing the fishery and keeping the history and knowledge of American shad going," says Kate Taylor, coordinator for the commission's shad plan.

Most shad sold in restaurants and stores this spring probably comes from Southeastern states with sustainable stocks, Taylor says. This year, she says, people should ask where the fish was caught. Next year you won't have to ask; all shad on the market will have to be harvested sustainably. "Shad is a great fish to eat, it's very healthy for you, and you're supporting a local fishery that's sustainable."

Shad, a large member of the herring family, are famously bony fish. Cooks get around that by baking the fish to soften the bones. But for most people, the real prize is the shad roe.

These eggs are delicate, and they're best in a saut? of butter. This week Fox is topping them with a sweet and sour bacon sauce and a side of saut?ed bitter greens to offset the sweetness. Roe is a bit of an acquired taste, he acknowledges, with "a liver-esque, innards kind of flavor. But it's a very smooth and rich flavor."

Customers seem to agree. "It's really selling very, very quickly," Fox says.

Other restaurants make shad and shad roe a spring tradition. If you were lucky enough to be at the Grand Central Oyster Bar in New York City today, you could dine on a shad filet and shad roe combo, served with the traditional bacon, for $29.95.

Fishmongers who weren't born yesterday say they've seen American shad journey from being a must-have seasonal item to something that only older people remembered to order. But Stanley Pearlman, for one, is glad to see shad coming back. "It's had a rekindled interest with the younger chefs looking for a product with history," says Pearlman, president of NAFCO, a seafood wholesaler in Jessup, Md. "It's rediscovering something that was lost."

Home cooks thinking to tackle roe should proceed with caution, Pearlman says. "You have to saut? it perfectly. If it's overcooked the grains become a little sandy. If it's done the right way, it's definitely a good thing to eat."

Cancer research targets a key cell protein

Blocking "don't destroy me" signals that normally sit on the surface of tumor cells and render them resistant to immune-cell attack slows the growth of a broad range of human cancers when they're implanted in mice, researchers have found.

The approach, reported by immunologists at the Stanford University School of Medicine, was effective against ovarian, breast, colon, bladder, liver, prostate and brain cancer cells. If the work can be repeated in people, the approach may someday help doctors marshal defender cells in patients' own bodies to fight cancers, the researchers said.

Key to the work is a cell protein called CD47, which is already being investigated in the treatment of leukemia.

CD47 sits on cell membranes and communicates with various immune cells, including macrophages, which gobble up foreign invaders in the body. It plays an important role in the normal life cycle of healthy red blood cells, telling macrophages to leave the cells alone.

In the study, the scientists injected the animals with antibodies that bind to CD47 and block out its protective signal.

"If we can block this signal, we can get the immune system to eat [the cancer cells] up," said Stephen Willingham, a postdoctoral researcher in the laboratory of immunologist Dr. Irving Weissman at Stanford and first author of a paper about the work.

The Stanford team examined cancer cells removed from patients with a variety of types of solid tumors. They found that CD47 studded the membranes of almost all of the cancer cells in their sample, suggesting that it is a molecule common to all cancers.

Placing the cells in lab dishes, the team administered an antibody: a protein that binds to CD47 and blocks it from warding off immune system cells. Macrophages ate the cells.

The researchers then implanted human tumor cells in mice for further study. They allowed the cancers to grow, and administered the antibody against CD47.

Antibody treatment inhibited the growth of almost all of the solid tumors and was able to wipe out some smaller cancers altogether, according to the report, which was published Monday in the journal Proceedings of the National Academy of Sciences.

Even when it didn't destroy a large tumor, Willingham said, the antibody prevented the cancer from spreading and wiped out metastases scattered through the body.

The therapy worked better in some cases than in others. One breast cancer case didn't respond to treatment at all.

"We never figured that out," Willingham said.

Researchers who were not involved in the study called it an important incremental step toward a novel cancer therapy.

Paired with surgery or other treatments, it could be used to stimulate a patient's immune system to fight cancer, said USC oncologist Dr. Heinz-Josef Lenz.

But scientists will have to evaluate the treatment's effectiveness and safety in humans.

Although the mice in the Stanford experiment didn't suffer severe side effects, that may not be the case in people. As with chemotherapy, "you run the risk of not only killing the tumor, but also the normal cells," said David DiGiusto, a researcher at City of Hope in Duarte who was not involved in the study.

Willingham said the only way to know how the antibody treatment will work in people will be to try it out. Plans to test the antibody therapy in leukemia patients in Britain are underway, funded by the California Institute of Regenerative Medicine. The team also hopes to test the treatment in patients with solid-tumor cancers within a couple of years, Willingham said.

The Stanford team certainly provided reason to suppose that therapies blocking CD47 could be important, DiGiusto said. They showed that patients whose cancers made a lot of CD47 fared more poorly than those whose cancers made less of it.

Advances in chemotherapy have dramatically improved the outlook for many cancer patients, but the side effects of this treatment are daunting. A new generation of chemotherapy drugs with fewer side effects is the goal of Edward J. Merino, assistant professor of chemistry at the University of Cincinnati.

Merino will discuss his efforts toward designing these new anticancer agents Tuesday, March 27, at The Chemistry of Life: Spring National Meeting and Exposition of the American Chemical Society in San Diego.

At that meeting, Merino will show a new anticancer agent that targets cancerous cells by zeroing-in on the "pollutants" produced by the characteristic metabolism within tumors. This agent, which Merino calls a "ROS-caged agent," is activated by oxidative stress, a signature of cancerous activity.

"Cancerous cells use energy differently from other cells," Merino said. "Like an automobile engine with the gas pedal pushed to the floor, they give off a unique sort of pollution because they are not as efficient in using energy. Cancer cells don't use metabolism all the way through. We have found key enzymes that contribute to this characteristic signature."

This signature results from the build-up of reactive oxygen species, known as ROS. This environment creates a chemical target that Merino and his team can target with chemical systems. Merino describes his findings as a preliminary description of the factors that can make drug delivery much more selective.

"We are looking for chemicals that operate like heat-seeking missiles that attack only cancerous cells," Merino said. "If we are successful, we can create drugs with far fewer side effects."

The side effects associated with chemotherapy hair loss and weight loss are generated from the mechanism used to target today's family of drugs. Because cancerous cells reproduce rapidly, chemotherapy targets rapidly reproducing cells. The problem, Merino said, is that cells in the stomach lining and cells in our hair follicles also reproduce rapidly so conventional chemotherapy kills those cells, too.

A better understanding of cancer metabolism is an essential part of the work, but so is the basic chemistry involved in creating targeted compounds.

"We are continually making new compounds," Merino said, "but very few of them give any indication of working. Out of hundreds of candidates, we have one moving into a mouse model, and another that looks promising."
